Description

Establishing a reproducible patient-derived colon cancer organoid culture can be difficult when freshly isolated tumor tissue contains variable cell populations, residual blood cells, incompletely digested tissue, or samples with inconsistent growth potential. Researchers may also need to prepare multiple growth factors separately, optimize their concentrations, and repeat culture adjustments before obtaining stable colorectal cancer organoids for downstream studies.

Biofargo Human Colon Cancer Organoid Culture Kit is a two-component culture medium system developed for the establishment and expansion of human colon cancer organoids. The kit combines 96 mL of Human Colon Cancer Organoid Basal Medium A with 4 mL of Human Colon Cancer Organoid Culture Supplement B to prepare 100 mL of complete organoid culture medium.

The complete medium supports three-dimensional culture of tumor cells isolated from human colon cancer tissue. The resulting human CRC organoid cultures can be used to observe tumor cell growth and proliferation and to investigate changes in tumor organoid response under drug treatment conditions.

By integrating the required culture components into a ready-to-mix format, the kit reduces the need to prepare and optimize multiple individual factors. It provides researchers with a more standardized workflow for colon cancer organoid culture, colorectal cancer disease modeling, drug screening, drug toxicity evaluation, and related tumor biology research.

Recommended Culture Workflow

1. Tumor Tissue Collection and Temporary Storage

Place freshly collected human colon tumor tissue in pre-cooled primary tissue preservation solution and keep the sample temporarily at 4°C before processing.

2. Tissue Washing

Under sterile conditions, transfer the tissue to a culture dish and rinse it with pre-cooled PBS. Repeat the washing procedure at least three times to remove residual blood and loosely associated material.

3. Tissue Mincing

Transfer the washed tissue to a 1.5 mL centrifuge tube and cut it into approximately 1–3 mm³ tissue fragments using sterile surgical scissors.

4. Tissue Digestion

Transfer the tissue fragments to a 15 mL centrifuge tube and add 10 mL of primary tissue digestion solution. Digest at 37°C for approximately 15–45 minutes. The required digestion time may vary according to tissue source, tissue size, and sample condition.

During digestion, gently pipette or agitate the tissue approximately every 10 minutes while monitoring the degree of tissue dissociation.

5. Filtration and Cell Collection

After digestion, add an appropriate stop solution and pass the tissue suspension through a 100 μm cell strainer to remove undigested tissue fragments. Centrifuge the filtrate and collect the cell pellet.

6. Optional Red Blood Cell Lysis

If substantial red blood cell contamination is visible, add approximately 1–2 mL of red blood cell lysis solution to the pellet and incubate at room temperature for about 2 minutes. Stop the lysis reaction, centrifuge the sample, discard the supernatant, and retain the collected cells.

7. Extracellular Matrix Embedding

Resuspend the collected cells in a suitable extracellular matrix at a recommended density of approximately 1 × 10⁵ to 1 × 10⁶ cells/mL. Dispense the cell-matrix suspension into a 24-well culture plate while minimizing bubble formation.

8. Matrix Polymerization and Culture

Incubate the culture plate at 37°C with 5% CO₂ for approximately 15–25 minutes to allow the matrix droplets to solidify. After polymerization, carefully add approximately 700 μL of complete human colon cancer organoid culture medium around each matrix droplet and return the plate to the incubator.

Important: Tissue dissociation conditions may require adjustment according to the origin, size, condition, and cellularity of each tumor sample.

Applications

Human colon cancer organoids provide a three-dimensional research model for studying colorectal tumor biology under controlled in vitro conditions. Because primary tumor samples can vary substantially in tissue composition and growth behavior, a standardized colon cancer organoid culture medium can help researchers establish more consistent workflows for model generation and downstream experimental analysis.

Human colon cancer organoid establishment: Culture tumor-derived cells isolated from human colon cancer tissue in a three-dimensional extracellular matrix environment.
Colorectal cancer organoid expansion: Expand established CRC organoid cultures for serial passaging and downstream experimental use.
Patient-derived tumor organoid research: Generate patient-derived colon cancer organoid models for studying sample-specific tumor characteristics.
Cancer drug screening: Evaluate changes in colorectal tumor organoid growth after exposure to candidate compounds or experimental treatment conditions.
Drug sensitivity testing: Compare organoid responses to different compounds, concentrations, or treatment schedules in research workflows.
Drug toxicity evaluation: Observe treatment-associated changes in organoid morphology, growth, and proliferation under defined experimental conditions.
Colorectal cancer disease modeling: Use human CRC organoids as an in vitro model for investigating tumor development and cancer-associated cellular behavior.
Tumor biology research: Study colon cancer cell growth, proliferation, tissue organization, and treatment response in a 3D culture system.
Preclinical cancer research: Incorporate human tumor organoids into exploratory drug discovery and translational research workflows.
Precision oncology research: Investigate sample-dependent treatment responses using patient-derived CRC organoid cultures for research purposes.

Why Use a Human Colon Cancer Organoid Model?

Traditional two-dimensional cancer cell cultures are convenient for routine screening but may not fully reproduce the three-dimensional organization and growth behavior of tumor-derived cells. Human colon cancer organoids are cultured within an extracellular matrix, allowing tumor cells to form organized three-dimensional structures that can be monitored over time.

This culture format provides a useful research platform for observing tumor organoid formation, growth, proliferation, serial expansion, and response to drug treatment. Patient-derived colorectal cancer organoids may also help researchers compare biological differences between individual tumor samples in cancer disease modeling and precision oncology research.

Biofargo Human Colon Cancer Organoid Culture Kit provides the basal medium and culture supplement required to prepare a complete medium for these workflows. Its two-component design simplifies medium preparation and supports more standardized handling across repeated experiments.
