Description

Establishing patient-derived cholangiocarcinoma organoids from primary tumor tissue can be technically demanding. Variable tissue quality, incomplete tissue digestion, inconsistent culture conditions, and limited post-isolation expansion may reduce organoid establishment efficiency and make long-term experiments difficult to reproduce.

Biofargo Human Cholangiocarcinoma Organoid Culture Kit is a complete in vitro culture system developed for the establishment, maintenance, and passaging of human cholangiocarcinoma organoids. The culture environment is designed to support three-dimensional growth while helping preserve relevant pathological and biological characteristics of the original tumor tissue.

This cholangiocarcinoma organoid medium is suitable for researchers constructing patient-derived cholangiocarcinoma organoid models from freshly collected tumor specimens. It supports the culture of multiple cholangiocarcinoma subtypes and provides a practical platform for tumor biology research, cancer disease modeling, drug screening, and drug sensitivity testing.

The two-component format combines 96 mL of Human Cholangiocarcinoma Organoid Basal Medium A with 4 mL of Human Cholangiocarcinoma Organoid Culture Supplement B to prepare 100 mL of complete medium. The integrated formulation reduces the need to source and optimize multiple individual culture factors.

Recommended Culture Workflow

1. Primary Tumor Tissue Collection

Immerse freshly collected cholangiocarcinoma tissue in pre-chilled primary tissue preservation solution and store temporarily at 4°C. Process the tissue as soon as practical to help preserve viable tumor cells.

2. Tissue Washing

Under aseptic conditions, transfer the tissue to a culture dish and wash with pre-chilled PBS at least three times to reduce residual blood and tissue debris.

3. Mechanical Mincing

Transfer the washed tissue to a sterile 1.5 mL centrifuge tube and cut it into approximately 1–3 mm³ fragments using sterile surgical scissors.

4. Enzymatic Digestion

Transfer the tissue fragments to a 15 mL centrifuge tube and add 10 mL of primary tissue digestion solution. Digest at 37°C for approximately 15–45 minutes, depending on tissue source, size, and digestion progress. Gently pipette or agitate every 10 minutes while monitoring the tissue.

5. Filtration and Cell Collection

Stop digestion with an appropriate neutralization solution. Pass the tissue suspension through a 100 μm cell strainer to remove undigested fragments, and centrifuge to collect the cell pellet.

6. Optional Red Blood Cell Lysis

When the collected pellet contains substantial red blood cell contamination, resuspend it in 1–2 mL of red blood cell lysis solution and incubate for approximately 2 minutes at room temperature. Stop the lysis, centrifuge, discard the supernatant, and retain the cell pellet.

7. Extracellular Matrix Embedding

Resuspend the isolated cells in an appropriate extracellular matrix at a recommended density of 1 × 10⁵ to 1 × 10⁶ cells/mL. Dispense the cell–matrix mixture into a 24-well plate while avoiding bubbles. Incubate at 37°C for approximately 15–25 minutes to allow the matrix domes to solidify.

8. Organoid Culture

After the matrix has solidified, carefully add 700 μL of complete human cholangiocarcinoma organoid medium around each matrix dome. Incubate the cultures at 37°C in a humidified atmosphere containing 5% CO₂.

Why Use a Cholangiocarcinoma Organoid Model?

Conventional two-dimensional cancer cell lines are convenient for routine experiments but may not adequately reproduce the three-dimensional organization and biological diversity of primary cholangiocarcinoma tissue. Patient-derived organoids provide a complementary model that can retain clinically relevant tumor characteristics while remaining accessible to controlled in vitro experimentation.

A cholangiocarcinoma organoid model can therefore support studies that require patient-specific tumor material, repeated sampling, long-term expansion, or parallel testing of multiple experimental conditions. When established successfully, the organoids can be used to connect primary tumor biology with downstream drug-response, biomarker, and disease-modeling workflows.

Why Choose Biofargo Human Cholangiocarcinoma Organoid Culture Kit?

Primary cholangiocarcinoma specimens can differ substantially in cellularity, fibrosis, necrosis, and previous treatment exposure. These factors may influence tissue digestion, organoid formation, and subsequent expansion. A dedicated culture system helps reduce the additional variability introduced by assembling multiple independently sourced culture components.

Biofargo Human Cholangiocarcinoma Organoid Culture Kit provides a ready-to-prepare, two-component medium designed specifically for human cholangiocarcinoma organoid culture. The formulation supports establishment, maintenance, and passaging within a consistent workflow, allowing researchers to focus on tissue quality, matrix handling, and experimental design rather than repeated medium optimization.
